对乙酰氨基酚(APAP)过量是药物性肝损伤(DILI)的主要原因。但是目前肠道微生物群和相关代谢物对APAP和肝功能的影响仍不清楚。最近,南方医科大学团队在期刊《Cell Host & Microbe》(IF=31.316)上发表题为“Liberation of daidzein by gut microbial β-galactosidase suppresses acetaminophen-induced hepatotoxicity in mice.”的文章,该篇研究发现APAP紊乱与独特的肠道微生物群落有关,即阴道乳酸杆菌明显减少。由于细菌β-半乳糖苷酶从饮食中释放异黄酮大豆苷元,接受阴道毛滴虫的小鼠显示出对APAP肝毒性的抗性。用β-半乳糖苷酶抑制剂消除了阴道毛滴虫对APAP暴露的无菌小鼠的肝保护作用。类似地,β-半乳糖苷酶缺陷型阴道毛滴虫在APAP处理的小鼠中比野生型小鼠产生更差的结果,但是这些差异通过大豆苷元给药得以克服。大豆黄酮阻止了铁死亡,这与法尼二磷酸合酶(Fdps)表达下降有关,该合酶激活了涉及AKT-GSK3β-Nrf2的关键铁死亡途径。该研究证明了阴道乳酸杆菌β-半乳糖苷酶释放大豆苷元抑制了Fdps介导的肝细胞铁死亡,为药物性肝损伤提供了新的治疗方法。
